Tannin-rich herbs are classic cooling, drying remedies, valued for supporting mucosal barrier integrity, calming weepy tissues, and providing topical antimicrobial effects.

Overview:
Tannins contribute astringent, barrier-supportive, antimicrobial, antioxidant, anti-inflammatory, and hemostatic actions. Through protein binding, they tighten lax tissues, reduce exudation, and form a protective film over skin and mucosa. They also inhibit microbial adhesion and modulate inflammatory signaling pathways such as NF-κB, while supporting antioxidant defenses including Nrf2 activation. Clinically, tannin-rich herbs are especially relevant for oral and throat care, acute diarrhea, hemorrhoids, urinary tract support, weeping dermatological conditions, and topical wound care.

Structures & Subcategories:
Tannins are larger polyphenols with multiple hydroxylated aromatic rings capable of strong interactions with proteins and alkaloids. They fall into two primary subgroups:

• Hydrolyzable tannins (HT): gallotannins and ellagitannins contain gallic or ellagic acid units esterified to a central sugar; inside the body, they hydrolyze to release active phenolic acids.
• Condensed tannins: also known as proanthocyanidins (PAC) and their smaller oligomeric forms (OPC), constructed from flavan-3-ol units such as catechin and epicatechin. 

While not true tannins structurally, related polyphenols such as the catechin gallate esters and theaflavins in Tea (Camellia sinensis) contribute similar astringent effects and are often grouped functionally as “tea tannins.”

Polarity, Solubility & Extraction:
Tannins are polar constituents, highly soluble in water (especially hot water). They extract well into infusions and decoctions, as well as low- to mid-range hydroethanolic tinctures (~20–50% EtOH). Higher ethanol concentrations reduce extraction of larger tannins and may promote precipitation. Tannins are poorly extracted into nonpolar solvents such as fixed oils or CO₂. Hydrolyzable tannins are susceptible to hydrolysis, while condensed tannins can gradually oxidize; protection from heat, light, and oxygen supports stability.

Pharmacological Actions:
Tannins are known for broad effects on epithelial barrier integrity, inflammation, microbial ecology, and the cellular response to oxidative stress. Their protein-binding activity underlies their classic astringent effects: tightening tissues, reducing secretions, forming a protective surface film, and supporting local hemostasis. Many tannins also inhibit microbial adhesion and biofilm formation, modulate inflammatory pathways such as NF-κB, support antioxidant defenses through Nrf2-linked pathways, and mildly inhibit carbohydrate-digesting enzymes. Microbiota-derived metabolites, including urolithins from ellagitannins, extend tannins’ effects beyond local tissue interactions to include systemic activity.
